Sekelompok 15 siswa dari SMA Elit dikumpulkan di SMA Kelas Atas yang sangat istimewa, untuk lulus dari SMA artinya berhasil bertahan hidup, tapi untuk lulus sangatlah sulit. Sekolah ini dipimpin oleh beruang yang disebut Monokuma, dan Monokuma menjelaskan bahwa untuk lulus mereka harus saling membunuh. Satu-satunya cara untuk lulus adalah membunuh salah satu teman sekelas dan pergi setelah membunuhnya, jika teman sekelas lainnya menemukan identitas si pembunuh, pembunuh akan langsung dieksekusi. Namun jika mereka gagal menangkap si pembunuh, hanya pembunuh yang akan diluluskan dan yang lainnya akan dimusnahkan.
